The structure of four new MT (metallothionein) genes of Tetrahymena thermophila and T. pigmentosa were characterized. The MT-2 genes from the two species are very similar, differing by 10 out of 2259 sequenced nucleotides, and the deduced amino acid sequences are identical. The MT-1 genes from T. pigmentosa and T. thermophila are also very similar, differing only by 3 nucleotides in the 5′-UT region. The promoter regions contain a TATA box and many stretches partially matching some regulatory elements such as metal-responsive (MREs), antioxidant-responsive (AREs), a CAAT box, a G-box, and AP1 and ACE-1 binding sites. The related coding and amino acid sequences were compared with those previously sequenced in Tetrahymena. This analysis revealed two independent events of duplication occurring in Cd- (MT-1 and MTT1) and Cu- (MT-2) induced MTs. This evolutionary pathway also explains the unusual length of these proteins, which are much longer than many MTs studied so far. Additionally, the orthology and paralogy relationships of the various MTs are presented. Finally, on the basis of phylogenetic analyses of Tetrahymena MTs, two evolutionary hypotheses are proposed. 相似文献

Trophozoites of Troglocorys cava were detected in all but one of the wild chimpanzee populations from Rubondo Island (Tanzania), with a prevalence ranging between 20% and 78%. However, the ciliate was absent in all captive groups. This revision of the classification of eukaryotes, which updates that of Adl et al. [J. Eukaryot. Microbiol. 52 (2005) 399], retains an emphasis on the protists and incorporates changes since 2005 that have resolved nodes and branches in phylogenetic trees. Whereas the previous revision was successful in re‐introducing name stability to the classification, this revision provides a classification for lineages that were then still unresolved. The supergroups have withstood phylogenetic hypothesis testing with some modifications, but despite some progress, problematic nodes at the base of the eukaryotic tree still remain to be statistically resolved. Looking forward, subsequent transformations to our understanding of the diversity of life will be from the discovery of novel lineages in previously under‐sampled areas and from environmental genomic information. 相似文献

The nucleolar organization in ciliate Didinium nasutum somatic interphase nuclei was studied using serial ultrathin sections and compared for various physiological states of the cell, namely, fed ciliates, starved ciliates, and dormant cysts. It has been shown that the interphase nucleoli are large structures with a complex architecture: the fibrillar component forms an intricate network in the macronucleus space, while the granular component is located inside this network. The structures looking as individual nucleoli in single sections are actually parts of branched nucleolar networks. The intricate nucleolar networks do not disintegrate after a 30-h starvation; however, the granular component becomes denser and develops numerous cavities filled with fine fibrils of a nonribonucleoprotein nature. In fed D. nasutum, the fibrillar structures on the periphery of nucleoli contain numerous pores (virtually absent in starved cell nucleoli), which can potentially serve for transporting newly synthesized rRNP. Branched nucleolar networks are undetectable in cysts. Their nucleoli are individual structures consisting mainly of the fibrogranular component. 相似文献

2004年8月-2007年3月,用活体观察和固定染色方法对桃花沟森林公园及符家滩土壤纤毛虫的群落特征进行了研究.结果表明:在桃花沟森林公园的土壤中共鉴定出纤毛虫75种,隶属于3纲11目29科40属,其中桃花沟景区的土壤纤毛虫有43种,普渡峡景区有51种;受人为干扰较为严重的符家滩土壤中有纤毛虫28种;在桃花沟森林公园土壤纤毛虫群落中,下毛目、肾形目和前12目为其优势类群;齿脊肾形虫、长圆膜袋虫、皮速体虫、活泼状似片尾虫为其优势种;桃花沟景区、普渡峡景区和符家滩的土壤纤毛虫Gleason-Margalef物种多样性指数分别为4.61、5.64、3.28;桃花沟景区与普渡峡景区土壤纤毛虫的群落Jaccard相似系数为0.25,桃花沟景区、普渡峡景区与符家滩土壤纤毛虫的群落Jaccard相似系数分别为0.22、0.18.表明自然环境与受人为干扰的环境中土壤纤毛虫物种多样性和群落结构有显著差异. 相似文献

In Paramecium, several kinds of the oral networks of fine filaments are defined at the ultrastructural level. Using the sodium chloride-treated oral apparatus of Paramecium as an antigen to produce monoclonal antibodies, we have begun to identify the proteins constituting these networks. Immunoblotting showed that all positive antibodies were directed against three bands (70-, 75-and 83-kD), which corresponded to quantitatively minor components of the antigen; there was no antibody specific for the quantitatively major components (58- and 62-kD). Immunolocalization with four of these antibodies directed against one or several of these three bands showed that these proteins are components of the fine filaments supporting the oral area; a decoration of the basal bodies and the outer lattice was also observed on the cortex. Immunofluorescence on interphase cells suggested that the three proteins colocalized on the left side of the oral apparatus, whereas only the 70-kD band was detected on the right side. During division, the antigens of the antibodies were detected at different stages after oral basal body assembly. The antibodies cross-reacted with the tetrins, which are oral filament-forming proteins in Tetrahymena, demonstrating that tetrin-related proteins are quantitatively minor components of the oral and the somatic cytoskeleton of Paramecium. 相似文献

Recent works on prostomatid ciliates show that some genera of this group have a differentiated oral infraciliature and that their stomatogenesis during division involves the proliferation of only a few somatic kineties. These findings have significant implications regarding the iaxonomic status of these genera and also on the terminology used for the oral structures. In Urotricha ondina , the oral infraciliature consists of (1) a paroral kinety formed of paired kinetosomes that encircle the cytostome at the anterior pole of the cell and (2) 3 adoral organelles, each formed of 2 rows of kinetosomes, ventral in position and obliquely disposed, lying above 3 short somatic kineties that do not reach the anterior pole of the cell. This oral ciliature —formerly known as the corona and brosse, respectively—originate during stomatogenesis from the proliferation of 4 somatic kineties that lie posterior to the adoral organelles of the parental cell. 相似文献

The species richness and seasonal development of planktonic ciliates were studied and compared in two shallow mesotrophic lakes, one covered with dense submerged macrophytes, the other macrophyte poor. Considerable differences in ciliate species composition, dominant taxa, abundance and biomass were observed. Ciliates were much more species rich in the macrophyte-rich lake, while they were more abundant numerically in the macrophyte-poor lake. Altogether, 96 species, included in 53 genera, 14 orders were identified. Among them, 80 species (included in 45 genera, 14 orders) observed from the macrophyte-rich lake, against 49 species (36 genera, 12 orders) were from the macrophyte-poor lake. In the macrophyte-rich lake, the mean abundance and biomass were 13.5 cells ml-1 and 547.10 g l-1 f.w.; abundance and biomass were higher in spring and winter; naked oligotrichs dominated total ciliate abundance and Peritrichida dominated the biomass. In the macrophyte-poor lake, ciliate mean abundance and biomass were 35.5 cells ml-1 and 953.39 g l-1 f.w.; abundances peaked in autumn; Scuticociliates dominated the abundance and Tintinnids dominated the biomass.
